<br/>Nanoparticle based therapeutics have great potential in the development of personalized medicine, however, the intracellular processing of such materials is difficult to actively monitor. Optical probe methods can offer great advantage for monitoring the uptake, encapsulation, and correlated cellular response to a delivered nucleic acid cargo on a gold or semiconductor quantum dot nanotheraputic. In this presentation we will demonstrate the use of a set of nano-gold bio-optical transponders (nano-BOT) capable of actively reporting the nanoparticle location, the timing of intracellular DNA cargo release from the gold nanoparticle (AuNP), the chemical environment of the delivered cargo, and the correlated cellular response to the therapeutic agent. In the presentation, we will discuss the design of multi-color surface energy transfer (SET) methods based on the chemical coupling of nucleic acids selectively to the surface of 7 nm gold nanoparticles, as well as explore peptide meditated nanoparticle uptake and delivery of therapeutic nucleic acids using live-cell imaging techniques. The presentation demonstrates the advantages of nano-BOT techniques for cellular biology questions.
