Residual pesticides in food and agricultural products have been recognized to influence on estrogenic (or androgenic) activity through various metabolism in human body. However, no systemic comparison on the metabolomic variation in response to estrogenic pesticides to date. In this study, we attempted to establish simple method to evaluate and identify active metabolites from estrogenic pesticides through metabolomic analysis of liver S9 fraction as <i>in vitro</i> hepatic model. 10 pesticides (pendimethalin, fenhexamid, methiocarb, chlorpropham, cafenstrole, bitertanol, tebufenpyrad, fenarimol, pyrimethanil and diazinon) which have estrogen receptor agonistic activity were incubated with liver S9 fraction then their metabolomes were analyzed with LC-MS/MS. Acquired data set were processed using MZmine2 and metabolomic variations were compared through multivariate statistical analysis (OPLS-DA, orthogonal partial least square discriminant analysis). OPLS-DA revealed significant metabolomic variation by enzymatic reaction of S9 fraction and several key metabolites were identified in 6 pesticides. Oxidized metabolites were identified in cafenstrole, bitertanol, pyrimethanil, and diazinon, while sulfoxidized compounds were screened in methiocarb metabolites. In addition, tebufenpyrad was oxidized as well as glycosylated through S9 fraction model. Estrogenic pesticides were primarily metabolized by oxidation in this hepatic model and some of key contributing metabolites were successfully identified by metabolomic analysis which would be useful tools for further application.
