Nanoparticles is a common carrier system to enhance systemic availability of bioactive compounds. Even though many ways to prepare nanoparticles, disadvantages, such as complicated processing procedure, and difficulties to harvest particles from solution are remained. The innovation technique, coaxial electrospraying, could overcome those struggles through one-step preparation. Thus, the objective of this work is to develop a convenient methodology to encapsulated curcumin into bilayer nanoparticle that could contain, protect, and enhance its absorption in gastrointestinal (GI) tract. Curcumin, although considered as a salubrious compound, their poor aqueous solubility and fast degradation rate in GI environment leads to low oral bioavailability. Therefore, being encapsulated in a delivery vehicle could significantly enhance their applicability in functional food product. The core material, chitosan, has positively charge that could enhance absorption rate with the negative charged intestinal lining. However, chitosan may dissociate in lower pH environment urges the need of additional protection layer. In this sense, negatively charge alginate are selected for fabrication of surface protection coating. To optimize properties of bilayer nanoparticle, RSM was used to find suitable processing parameters and compositional ratio of curcumin and chitosan. The encapsulation efficiency, particle size, conductivity and viscosity of those polymer solutions are measured by UV-vis spectroscopy, dynamic light scattering, conductivity meter and rheometer, respectively. In addition, the morphology of bilayer nanoparticles was studied using transmission electron microscopy. The conductivity and viscosity of chosen formulation was set between 1.03-8.84 mS/cm and 0.01-1.39 Pa.s, respectively, suitable for producing nanoparticle in electrospraying. The optimize curcumin containing nanoparticle had particle size around 165 nm and surface charge at -32 mV. The formation of bilayer structure confirmed by microscopic observation indicated by distinct two-layer structure. Moreover, the encapsulation efficiency of curcumin in bilayer nanoparticle was above 90% indicating low processing loss of bioactive component. To sum up, coaxial electrospray is an efficient method is to produce multi-layered carriers in one-step that could save times and production costs. This technique can definitely replace complex process of constructing delicate delivery system for bioactive compounds.
