Electrophilic molecules from natural products to lipid electrophiles induce the cytoprotective heat shock response (HSR). The molecular mechanisms that undergird this induction involve heat shock protein 70 (Hsp70), Hsp90, and the heat shock transcription factor 1 (Hsf1). The effects of various electrophiles on the Hsf1 mediated HSR induction have been characterized through <i>in vitro </i>electrophoretic mobility shift assays and transcriptional activation assays. Small molecule capture of Hsp90 allows identification of electrophilic modification sites on the protein after treatment of human leukemia cells with electrophiles such as parthenolide.
