While the notion of an “ideal synthesis” in the context of complex secondary metabolites was described as early as the 1970s, the application of these same principles to the molecules of life remains largely underexplored. This discussion will highlight our efforts toward ideality in biomolecule construction with an eye toward (1) minimization of concession steps and (2) streamlining the standard iterative processes currently employed. Similarly, where non-natural functionalization is desired, direct modification of native sequences—with an emphasis on complete chemoselectivity and precise stereocontrol—will be detailed.
