The pyridoxal and hydrazide based schiff bases are known to show wide range of biological activities 1,2,3 as antimicrobial, antiviral and anticancer and can also act as good chelating agents. The transitional metal chelates of these ligands have been reported to show significant biological activities 4,5. Accordingly, the transition metal complexes of M= Cu(II) and Zn(II) involving Schiff base ligand, L=4-hydroxy-N-((3-hydroxy-5-(hydroxymethyl)-2-methylpyridin-4- yl)methylene)benzohydrazide have been synthesized and characterized by elemental analysis, magnetic susceptibility measurements, molar conductance, IR, 1H-NMR, TGA, UV, LC-MS and ESR spectral studies. The results suggest that the ligand acts as tridentate and forms ML2 (1:2) complexes with octahedral geometry. Their electronic absorption, fluorescence emission and viscosity measurements were carried out to study the DNA binding activity of the metal chelates and found that the complexes bound to Calf thymus DNA in an intercalative mode. The plasmid pBR322 DNA was used to study DNA cleavage activity of complexes wherein the complexes showed good cleavage activity. In addition, the antibacterial studies were also carried out and the metal complexes were found to show better activity than the ligand.
