The Dutton-Discher laboratory has engineered simple, artificial proteins that facilitate electron transfer, termed ‘maquettes’. This maquette technology is currently being utilized to develop novel genetically-encoded voltage indicators (GEVIs), which optically report changes in membrane potential. Site-directed mutagenesis was completed on a particular water-soluble maquette (MZH3) to redesign the maquette into an improved potential GEVI with the capability of being cross-linked to a nanoparticle. This new construct may then be further modified such that it has the ability to report voltage by electron transfer-dependent fluorescence quenching of this tethered nanoparticle.
