Heparin is an anticoagulant commonly used to prevent postoperative thrombosis. Because the complications of heparin include thrombocytopenia, bleeding, and hemorrhaging, accurately measuring the concentration of heparin and administering the correct amount of its antidote is critical. While there are optical and electrochemical methods currently used to measure heparin, their instrumentation limits point-of-care testing and in some cases involves an indirect measure of heparin. This study investigates the development of a paper-based analytical device (PAD) to directly determine heparin concentrations. This PAD exploits chromoionophore-based sensing for measuring heparin utilizing a smartphone app as the detector. This presentation will describe the creation of the PAD and its optimal composition, including the different ratios of the ion-exchanger tridodecylmethylammonium chloride and Chromoionophore IV used, as well as construction of a lightbox that enables consistent measurements. Additionally, the optimal elapsed time between exposure to heparin and hue measurements, and the ability to measure heparin in plasma will be reported.
