We have developed a new ion-pair high-performance liquid chromatographic method for the determination of biologically significant aminothiols. The method utilizes a post-column reaction of each aminothiol with a 2,4-dinitrobenzenesulfonate derivative of a 3-hydroxyflavone followed by fluorescence detection. Accuracy, precision, post-column temperature and residence time optimization, and limit-of-detection are evaluated and results are compared to an existing HPLC method coupled with pre-column derivatization for homocysteine, cysteine, cysteinyl-glycine, and glutathione. Improvement in sample throughput and reduced sample preparation time over existing methods is also assessed and presented.<br/>
