G-quadruplex (GQ) with some interesting applications, including supramolecular hydrogel, nanomaterials, potential targets for cancer therapy, has been developed extensively over the past decades.  However, there is still a challenge in developing novel supermolecule with high stability in polar protic solvent.  One general approach to achieve structurally rigid G-monomer is the modification on guanine (8-aryl) and ribose (2’, 3’-isopropylidene).  According to ion dipole interaction between metal cations and central oxygen atoms, stacking of G-quartets formed by H-bond gives G-quadruplexes.  A novel system with enhanced stability in methanol and DMSO were developed via cross layer H-bond and covalent linkage. The structure of the guanosine octamers were characterized by X-ray single crystal diffraction, MS and NMR.<br/>
