In order to combat the growing threat of antibiotic resistant bacteria, new antibiotics must be developed. While natural products can be used as scaffolds, the acquisition of large amounts of these natural products can prove arduous and costly.  Total synthesis in the laboratory provides access to these natural products and means for producing enough material for further study. The family of compounds called Calopins has recently been reported to have activity against antibiotic-resistant bacteria. However, only the simplest known Calopin has been synthesized. Specifically, the synthesis of the complex base structure of recently discovered Calopins, which consists of multiple rings and up to seven stereocenters, has yet to be reported. This proposed total synthesis of Calopin 8-deacetylcyclocalopin B provides a new avenue for accessing the complex Calopin base and further exploring the bacterial resistance of this class of compounds.
