The site selective modification of carbohydrates and natural products is a fertile area for generating novel complex molecules, and an excellent testing ground for developing selectivity approaches within small molecule catalysis. Nature has demonstrated that substrate binding is an effective means of accelerating chemical transformation as well as controlling the selectivity. Herein, we will discuss the design of catalysts that leverage proximity effects to enhance the rate and site selectivity in the functionalization of complex molecules. Through the use of reversible covalent bonding to directly link the substrate to the catalyst, we were able to overturn the substrates inherent reactivity bias enabling the functionalization of the least reactive sites within polyol containing compounds.  <br/>
