<br/>Xanthene dyes are a group highly fluorescent compounds used in a wide variety of applications, including clothing dyes, fluorescent labels in biological assays, and as lasing media. Their significant absorption cross-sections and photostability have promise in light-harvesting applications. However, in concentrated solutions, or in dense materials, intermolecular interactions can complicate xanthene dye photophysics. To investigate how the spatial arrangement of fluorophores affect the photophysical properties of a xanthene dye, novel rhodamine B dimers were linked by 1,4- diamines with unique conformational stereochemistry and flexibility. The dimers were analyzed in differing solvents using fluorescence correlation spectroscopy, fluorometry, and ultraviolet–visible spectroscopy. Optical analysis of the dimers revealed a wealth of data, including absorption and emission maxima, hydrodynamic radii, molecular brightness, and processes occurring at sub-diffusion timescales. Molecular dynamic simulations were employed to model intradimer stereochemistry, such as π-π interactions. Across the rhodamine B species, the choice of solvent and type of linker strongly impacted their photophysical and solubility properties which potentially can guide the design of systems which employ xanthene dyes in higher density applications.
