Alzheimer’s disease (AD) is the leading cause of dementia, affecting about 5.5 million Americans. A key indicator of Alzheimer’s disease is an increase in amyloid beta (Aβ) plaques found within neuronal cell bodies in the brain, contributing to neuronal death.  This results in the slow loss of memory and motor function that accompany Alzheimer’s disease. Electron transport chain malfunction in mitochondria, an early indicator of AD, is linked to oxidative stress, as a result of reactive oxygen species (ROS). SH-SY5Y cybrid neuroblastoma cells provide a model for study of the role of mitochondria in the production of Aβ and the response to ROS in the AD neuron.  Carnosic acid (CA), a compound found in rosemary and sage leaves, may provide protection against Aβ as it triggers neuroprotective autophagy and decreases ROS. Treatment of SH-SY5Y cybrid cells with CA provides an opportunity to investigate the role of mitochondria in the neuroprotective effect of CA. SH-SY5Y cybrid cells lines will be screened for the presence of Aβ (1-42) using immunohistochemistry (IHC). The CA effect on ROS and Aβ toxicity in SH-SY5Y cybrid cells will be investigated using IHC and flow cytometry. <br/> <br/>
