This talk will discuss the vibrational modes characteristic to crystalline celluloses, the orientation of transitional dipole moments of selected vibrational modes with respect to the cellulose chain direction (c-axis), and how the concentration and spatial distribution of crystalline cellulose domains interspersed in amorphous matrices are manifested or analyzed differently in infrared, Raman, and sum frequency generation (SFG) vibrational spectroscopy analyses. This talk will highlight differences in physical principles among these three vibrational spectroscopic methods and demonstrate how such differences can be combined to obtain comprehensive information about the structure and crystallinity of cellulose in plant cell walls. Comparison with other crystallinity calculation methods will also be discussed.
