Exosomes are small in size (30 to 100 nm in diameter) extracellular vesicles containing in their membrane a rich variety of bioactive molecules, including receptors, ion-channels, membrane proteins, and various biomarkers depending on the type and state of the cell of origin. The exosomes have an extraordinary ability to transmit genomic and cellular state information between cells. Hence, they constitute an ideal medium for dependable cancer diagnostics and can also be utilized for an effective cancer therapy. Our investigations of exosomes are focused on glioblastomas, the most common and deadly tumors of the central nervous system. In this work, we have developed electrochemical and nanogravimetric immunosensors for the detection of exosomes isolated from glioblastoma U-87 MG cells. The proposed biosensor is based on the gold piezoelectrode modified with CD274 monoclonal antibody. The interactions of exosomes with nanoparticles functionalized with temozolomide anti-cancer drug have also been investigated. The effects of pH, glutathione concentration, and temperature on the controlled-release drug delivery have been analyzed.
