The research and development of new active pharmaceutical ingredients (APIs) has been complicated by issues with solubility and bioavailability. In order to solve this issue, APIs are often ionized into a salt form which improves the solubility. Currently, most salt APIs are in a solid form. However, since liquid salts, or ionic liquids, have been found to be biologically active, they make ideal candidates for pharmaceutical applications to be included within an API. This liquid salt form of an API or an Active Pharmaceutical Ingredient – Ionic Liquid (API-IL), combines the diversity and tenability of ionic liquids with the pharmaceutical use of APIs to unlock a new potential treatment option. API-ILs have the potential to deliver a drug by connecting the API to the ionic liquid using an ionic bond, covalent bond or both. This project focused on the synthesis and biological evaluation of Nonsteroidal anti-inflammatory drugs (NSAIDs) –based ionic liquids. NSAIDs were incorporated into pyrrolidine-based ionic liquids using ionic, covalent or both ionic and covalent links. Ionic-bound NSAID-ILs were prepared by using anion metathesis reactions. Covalent-bound NSAID-ILs were prepared using a two-step synthetic pathway. These ionic liquids can act as prodrugs for NSAIDs and should be hydrolyzed in the body to release the active form. All NSAID-ILs were characterized by spectroscopic methods. Based on our toxicity studies, new NSAID-ILs have shown only similar toxicity to regular NSAIDs. Efforts are underway to prepare dual-active API-IL drug delivery platforms containing two APIs.
