Published data supports the vital roles that gut microbiota play in the pathophysiology of obesity and related inflammatory conditions. Our group has previously reported on the biological effects, including anti-diabetic potential, of a phenolic-enriched red maple (Acer rubrum) leaves extract (MaplifaTM) standardized to glucitol-core containing gallotannins. Herein, we evaluated MaplifaTM effects on diet-induced obesity and gut dysbiosis in C57BL/6 mice fed either a standard diet (SD) or a high-fat diet (HFD) with or without MaplifaTM (150 mg/kg/day) by oral gavage for 8 weeks. In the HFD-fed animals, MaplifaTM supplementation significantly decreased weight gain, liver and kidney tissue weights, visceral obesity, hyperlipidemia and liver steatosis. MaplifaTM supplementation also restored the impaired glucose and insulin tolerance, improved insulin resistance and insulin signalling transduction in the HFD-fed mice. Additionally, MaplifaTM reduced white adipose tissue expansion and the inflammatory response in white adipose tissue. MaplifaTM supplementation modulated gut microbiota composition and certain bacteria population, including Bacteroides, Prevotella, and Clostridium XlVb as revealed by 16S rDNA analyses. Our work for the first time identified a novel causal linkage between gut microbiota alterations and metabolic improvement of MaplifaTM administration. The results of this study support the potential of MaplifaTM as a dietary ingredient for the prevention and/or treatment of obesity related metabolic diseases.
