Because of the small size of bacterial cells, the mysteries of their subcellular structure, dynamics and cooperativity are hard to understand with conventional approaches. We have been developing new single-molecule and super-resolution microscopy methods to locate, track, and analyze motion in living bacterial cells. I will outline some new methods that we have developed to isolate the foreground with accurate background subtraction, to unravel the complexities of heterogeneous motion with Bayesian statistics, and to measure very fast diffusion with image correlation. These techniques will be presented in the context of some of our recent findings about DNA replication in <i>Bacillus subtilis</i> and nutrient metabolism in the human gut microbiome.
