Gemcitabine and 6-Mercaptopurine (GEM and 6-MP) are wildly used chemotherapeutic agents clinically. However, as cytotoxic anticancer drugs, GEM and 6-MP's treatment outcomes are always accompanied with serious side effects. Here, a new luminescent sensor based on MOFs platform was prepared and shows high sensitive and selective determination toward GEM and 6-MP. An azamacrocyclic hexacarboxylate ligand and terbium cations were served to construct the water-stable metal-macrocyclic framework (MMCF-4) which exhibits a hexagonal honeycomb-type framework with an aperture of 15 Å. The carboxyl-rich ligand can not only form three binding sites per metal node, but also significantly enhance the stability of the MOF by protecting the Tb<sup>3+</sup> in a stable nine-coordinate tricapped trigonal prismatic environment. Besides, the tertiary amine on the ligand could redirect the analyte toward binding sites through base-base repulsion, thus further increasing the detection sensitivity. MMCF-4 displays excellent sensitivity with fast response. The detection limits can reach as low as 0.27μM and 0.89μM for GEM and 6-MP in aqueous media. The study introduces a Ln-MOF based sensor which can potentially serve as diagnostic tool for chemotherapeutic agents and a general route to reduce toxicity effects and improve drug efficacy for cancer patients.
