Individual and combined effects of nitrate (NO3-), metformin, and Adenosine Monophosphate-activated Protein Kinase (AMPK) inhibitor dorsomorphin (compound C) on the primary and cancer bladder cells are examined in this report. Viability profiles of primary bladder cell (PdEC) with concentrations of NaNO3 from 30 to 500 mM resulted from 75.7 to 0% cell viability in 24 h. Resulted cell viability data with concentrations of metformin from 1.0x103 to 1.0x104 mM of the same cell line are 98.7 to 95.9% in 48 h. Addition of both 300 mM NaNO3 and 1.0 x 104 mM metformin to PdEC resulted in 82.5 and 80.5% cell viabilities in 24 and 48 h, respectively, compared to 70.7 and 48.8% cell viabilities when treated individually with 300 mM of NaNO3 and 99.2 and 98.0% when treated with 1.0x104 mM of metformin alone within the same experiment and duration. Sequential treatments of nitrate, metformin and compound C in 72 h timeframe showed interesting results. Addition metformin first or later with respect to NaNO3, 24 h first with metformin and followed by 48 h with NaNO3 or 24 h first with NaNO3 and continued with 48 h of metformin, does not change the % cell viability at about 91.3 to 91.7%. However, pretreatment the PdEC with compound C for 6 h prior to the sequential treatments of both addition orders of NaNO3/metformin resulted in significant reduction of cell viability to 57.6 to 60.9%. The cell viability of PdEC treated with 1.0x104 mM of compound C alone was found to be at 96.4% after 72 h. The viability profiles of bladder cancer cell (5637) with metformin concentrations of 1.0x103, 5.0x103, and 1.0x104 mM were found to be at 89.1, 82.1, and 46.0%, respectively. Trypan blue exclusion assay method was used to determine the cell viability profiles in this study.
