It is estimated that 13.8 million Americans will be diagnosed with Alzheimer’s disease (AD) by 2050. As a result, a prolific amount of research is being conducted to identify possible targets for the treatment of AD. One recently identified target is the tau protein as increased levels of the aggregated form of tau have been found in patients with AD. As such, there has been an increase in research that focuses on identifying compounds that inhibit the self-aggregating properties of tau. Rhodanine structures have been found to strongly inhibit this property in the tau protein; however, they are toxic to the body by themselves. Flavonoids, a class of non-toxic, naturally occurring compounds that have been shown to exhibit various pharmacological activities, are of interest as potential inhibitors. By combining the rhodanine structure with the flavonoid compound, novel ligands were designed and evaluated for their binding affinity using molecular docking studies. Following which, synthetic pathways were proposed for the most promising compounds. Novel compounds were successfully synthesized, characterized, and will be evaluated for their pharmacological activity.<br/>
