Coenzyme A (CoASH), acting as an acyl carrier and co-substrate, is important for many enzyme reactions. However, the price of the commercial coenzyme is high (2,600 USD/g with &gt;85% purity) as the synthesis requires multiple reactions and purifications. CoASH could be produced by reducing CoA-S-S-CoA (CoA dimer) which is cost-effectively synthesized from chemically prepared pantethine through enzyme reactions (pantetheine kinase, phosphopantetheine adenyltransferase, and dephospho-coenzyme A kinase). However, the CoA dimer purification by using an anion exchange column from all enzyme reaction components is still laborious, and thus hampers scale-up. We tested different charcoals and solvents in an effort to remove the column purification step and make the CoA dimer preparation efficient.
