The Ugi-Smiles multicomponent coupling reaction utilizes amine, aldehyde, isocyanide, and phenol components in a highly atom-efficient, one-pot process to produce α-aminoamide products. In the presence of a conjugated aldehyde, a novel five-component Oxy-Michael Ugi-Smiles reaction was observed, with the alcoholic solvent participating in the reaction. Efforts are focused on exploring the reaction pathway and optimizing the observed five-component coupling through variations in the alcohol, amine, and aldehyde components.
