Our goal is to create covalently-linked fluorescent dye dimers using Rhodamine B as the dye and four different linkers that exhibit different fluorophore spacing, stereochemistry, and conformational flexibility. We hope to investigate the impact of fluorophore intramolecular association and relative geometry on the photophysical properties of the dimers. In this presentation, we will report the synthesis of these four fluorescent dimers with the various linkers and their synthetic characterization data. The optical properties of these different dimers will be quantified using a UV-Vis spectrophotometer, fluorometer, and fluorescence correlation spectroscopy (FCS), which measures temporal fluorescence fluctuations caused by single fluorescent molecules moving through an excitation volume. A more thorough understanding of the interplay between linker & fluorophore stereochemistry versus photophysical properties could inform the design of fluorescent poly-fluorophores, characterization of fluorophore aggregation, and relationship between inter-fluorophore geometry and energy transfer.
