Ions are agents of chemical synthesis as much as they are of analysis. The development of ambient ionization [1] makes increasingly accessible experiments in which ions are used for synthesis. The observation that reactions in small droplets or thin films are accelerated by orders of magnitude [2] makes possible the scale up of ion-based synthesis, while working in the conventional solution phase. This study explores the mechanism and the analytical and synthetic potential of high throughput reactions using the spray-based ambient ionization method of desorption electrospray ionization (DESI). By scanning the spray across a microarray at a rate of 1 spot/sec, thousands of reactions can be screened automatically for products at a rate of one reaction mixture per second [3]. An automated system that implements these capabilities is described. The mechanism of reaction acceleration is addressed with the aid of experiments on levitated droplets [4] and reaction acceleration in thin films [5].
