A longitudinal study regarding peptoid nanosheet stability was completed using fluorimetry and confocal microscopy techniques. Synthesis of 12-mer peptoids (Nae-Npe)<sub>6 </sub>– (Nce-Npe)<sub>6</sub>was accomplished using a solid-phase sub-monomer synthesis method. Aliquots of each peptoid solution were diluted with either PBS or Tris buffer and rotated on a vertical revolving plate to self-assemble into highly ordered nanosheet secondary structures. Fluorimetry data was obtained from each sample and resolved two peaks (612 nm and 650 nm) that were analyzed for the duration of this study. Analysis of the data showed no significant difference for peptoid nanosheet stability between PBS and Tris buffer as well as with each concentration of Nile Red dye (0.5 and 1.0 μM). Determining the relative stability of peptoid nanosheets with various conditions is significant to optimize the durability of the nanosheet structures and long-term surface adherence.
