The development of technology that can enable detection of hepatitis B virus (HBV) infection at the initial stage is important. Therefore, this study aimed to develop a more sensitive method of hepatitis B surface antigen (HBsAg) detection using gold nanoparticles (AuNPs). A significant increase in localized surface plasmon resonance (LSPR) signals was observed because of the electromagnetic amplification properties of AuNPs. As a result, an LSPR sensing chip was developed with antigen-antibody reaction-based detection symmetry, which detected HBsAg at 10 pg/mL. To further improve the detection limit, a modified detection format was created by fixing a secondary antibody to the AuNP monolayer. This enhanced the detection sensitivity by about 100 times. The developed LSPR sensing chip was able to detect as little as 100 fg/mL of HBsAg within 10-15 min. This is an improvement over a previously developed lateral flow assay and commercial diagnostic lateral flow kit, which can detect 10 ng/mL of HBsAg within 10 min. In addition, the LSPR sensing chip did not show any non-specific binding to other tested antigens, including alpha fetoprotein (AFP), C-reactive protein (CRP), and prostate-specific antigen (PSA). These findings confirm that the proposed detection strategy of LSPR sensing chips may provide a new platform for early diagnosis of various human diseases.
