The coordination sphere of paramagnetic transition metal complexes can be tuned to produce metal centers with different spin and oxidation states for applications as magnetic resonance imaging (MRI) contrast agents. For example, our laboratory has focused on Co(II) complexes that produce highly shifted proton resonances that are suitable for paramagnetic chemical exchange saturation transfer (paraCEST) agents and for water proton shift agents for cell-CEST. Imaging studies with a series of Co(II) macrocyclic complexes will be described. To produce new T1 relaxivity agents for MRI, high spin Fe(III) complexes have been prepared. Fe(III) complexes with innersphere water ligands or with only second sphere water ligands will be compared as T1 contrast agents. A few of these Fe(III) complexes are promising T1 agents for detection and characterization of murine tumors. The role of charge and functional groups of the complexes in modulating biodistribution and clearance in mice will be presented.
