In the past decade, a large number of green fluorescent proteins (GFP) based biosensors have been developed and catalyzed biological research in various of areas. However, compared to green fluorescent ones, red fluorescent biosensors are preferable because of lower phototoxicity and increased tissue penetrations. Furthermore, GFP based biosensors are not compatible with most existing optogenetics tools as they both require blue excitation light. Although recent studies have developed a few red fluorescent biosensors based on red fluorescent proteins (RFP), each required significant effort in optimization. We here present a general strategy to transfer a large number of GFP-based biosensors into red fluorescent biosensors.
