Isotopically labeled compounds are of high interest for several applications such as internal standards for mass spectrometry or as markers for mechanistic studies in organic and organometallic chemistry. In addition, they can be used to enhance the pharmacokinetic properties of some drugs in pharmacology. Here, we focus on the direct labeling of molecules with biological interest and more precisely on the direct isotopic exchange (H/D or H/T) using nanoparticles (NPs) as catalysts in water as the solvent. First we synthesized a new class of water soluble ruthenium nanoparticles. The NPs were stabilized by NHC (N-heterocyclic carbene) ligands, as this family of ligands provides a strong coordination to the NP surface. Another advantage of the NHC ligands is the possibility to modify their substitutents and thus to tune their physico-chemical properties. Sulfonated NHC ligands have been used to obtain water soluble ruthenium NPs. Unfortunately ruthenium NPs are very active in bond cleavage reactions and sometimes decompose or reduce the substrates. We decided to turn to alloys in an attempt to passivate the Ru surface. Thus, bimetallic Ru-Pt nanoparticles have been prepared in order to tune the chemical composition of the NP surface and thus influence the H/D exchange process, as platinum is not active in the H/D exchange. We thus synthesized different Ru-Pt NPs using three platinum precursors (Pt(NBE)3, Pt(CH3)2COD and Pt2(dba)3. Indeed we were able to modulate the reactivity and also the selectivity of the catalysts by modifying their surface metal composition.
