Pathogenic bacterial species can cause serious and life-threatening infections, more specifically actinobacteria infections can lead to eumycetoma (madura foot) which is a debilitating infection common in developing countries. Siderophores are key components in bacterial virulence and in the bacterial endogenous secondary metabolome, and as such provide a unique insight into bacterial infection and potential treatments.<br/>We have isolated the bacterial siderophore madurastatin C1 from an <i>Actinomadura</i> sp. DEM31376. We have revised the structure of madurastatin C1 though a combination of semi-synthesis and analytical chemistry. Based on this we have completed the fist total synthesis of madurastatin C1 via a convergent synthetic route utilising oxidised amino acid chemistry.
