Our project focuses on the synthesis of new complexes based on the active sites of superoxide dismutase (SOD) enzymes, which catalyze the disproportionation of superoxide to form dioxygen and hydrogen peroxide. The active sites of SOD enzymes all consist of first row transition metal centers, and many of the active sites contain imino and thiolate groups coordinated to the metal. We will present results of our efforts to synthesize iminopyridyl thiolates and N9-(2-ethanthiol)adenine (AdSH), as well as first row transition metal complexes that incorporate these ligands.<br/>We have shown that condensation of 2-acetylpyridine or 2-pyridinecarboxaldehyde with 2-aminoethanethiol hydrochloride results in formation of cationic thioaminal species. The thioaminals were deprotonated upon addition of two equivalents of base to form uncyclized iminopyridyl thiolates that react with cobalt and iron salts to form the corresponding metal complexes. When reacted with cobalt(II) chloride, AdSH forms a complex in which the ligand coordinates through an imino nitrogen atom in a 2:1 ligand to metal ratio. We will present characterization of these ligands and metal complexes. Additionally, we will present our results of electrochemical studies of the complexes and the viability of the new compounds to catalyze the disproportionation of superoxide.
