Pyruvate formate-lyase activating enzyme (PFL-AE) is a model radical S-adenosyl-L-methionine (SAM) enzyme due to its stability during purification and spectroscopic activity. However, PFL-AE has proven difficult to crystalize, with only two structures having been obtained in the past few decades and subsequent efforts to crystallize being unsuccessful. A computationally engineered PFL-AE with twelve surface mutations (PFL-AE-mut12) has been designed to improve crystallization for atomic-level structure determination.
