At Merck we aim to develop efficient, innovative, green and sustainable commercial synthetic routes to potential drug candidates using the best chemistry at time of regulatory filing, with an ultimate aspirational goal of the ‘zero waste’ API manufacturing process. A green by design development strategy allows progression from initial route design through to a fully optimized sustainable commercial manufacturing process. In many cases the discovery of new reactions/methodologies is required to realize these goals. This personal (34+ years at Merck) perspective, using selected case studies, will highlight the importance of synthetic chemistry, new technologies and innovative process optimization in the design and development of late stage API syntheses.
