Oxazoles are compounds with an array of biological applications primarily attributed to their molecular structure and substitution scheme. The ability to rapidly synthesize functionalized oxazoles with diverse substitution patterns is a robust area of study in synthetic organic chemistry. The overreaching objective of this research is to optimize the synthesis of diverse and highly substituted oxazole compounds from isocyanate and isonitrile starting materials, with a focus on improving the efficiency and regioselectivity of the reaction. Recently, we have synthesized tri-substituted oxazoles with an unprecedented 2,4-diamido-5-amino substitution arrangement. Using conventional heating, two identical amido groups can be added to the 2 and 4 position of the oxazole. However, two different amido groups can be added to the 2 and 4 positions by running the reaction in a microwave reactor, leading to a higher and more consistent yield. Our future goals are to continue exploring the scope and limitations of these reactions and continue expanding our library of newly synthesized oxazole compounds.
