Icetexane diterpenoides are highly complex natural products with a 6-7-6 tricyclic core structure and broad array of biological activity. Premnalatifolin A (1)—extracted from stem-bark of Indian plant, Premna-Latifolia—is a dimeric icetexane diterpenoid shows cytotoxic activity against MCF-7 cancer cell lines (IC50= 1.77 μM). We report here a general approach toward the synthesis of icetexanes core structure and synthesis of southern monomer of Premnalatifolin A as well as related analogues.
