Chlorobaculum tepidum, part of the family Chlorobiaceae, is an anaerobic, Gram-negative bacterium. As part of the family Chlorobiaceae, Cba. tepidumis a photolithoautotroph, using anoxygenic photosynthesis to fix CO2into biomass and provide energy for growth. It oxidizes sulfide to produce extracellular S(0) that is oxidized to sulfate after sulfide is consumed. Growth on S(0) requires biogenic S(0), i.e. the S(0) has to be synthesized by Cba. tepidumfor growth to occur. Proteomic analysis of biogenic S(0) indicates that it contains more than 100 proteins, many of which have been reported to be cargo of Outer Membrane Vesicles (OMVs). OMVs are shed by Gram-negative bacteria and are implicated in extracellular transport of cargo molecules. Data will be presented from dialysis cultivation experiments designed to test whether OMVs are involved in transport of S(0) to extracellular globules during the growth of Cba. tepidumon sulfide. This will begin to determine if OMVs are involved in S(0) metabolism of Cba. tepidum.
