Paper-based lateral flow strips, which is exemplified by the pregnancy test stripes are inexpensive and attractive in the point-of-care (POC) settings. However, it is usually used for qualitative analysis but for quantitative measurement of analytes. The reason is that gold nanoparticles are used for colorimetric measurement. In addition, the colorimetric paper test strips cannot be applied to blood samples due to severe interference from the sample matrix. To solve these problems, we have developed the sandwiched gold nanostar@Raman reporter@silica nanoparticles as the surface-enhanced Raman scattering (SERS) probes. This new type of SERS probes is characteristic of excellent water-solubility, high sensitivity and stability in blood samples. This SERS-based papers strip has demonstrated quantitative measurement of protein biomarker in the blood samples from clinical traumatic brain injury (TBI) patients.
