Silylated tag (STag) enabled peptides highly soluble in CPME recognized as a green solvent. Condensation reaction at high concentration in mixed solvent (CPME/DMF) has revealed to enhance reactivity and STagPS has achieved to synthesize even congested peptides in excellent purity. In addition, effective work-up procedure to remove a little excess of activated ester or dibenzofluvene from reaction mixture, allowed peptide elongation to be highly practical in terms of speed and scalability.
