Thiols play significant roles in biological systems. A status change of thiols has been observed in various diseases such as Parkinson’s disease and Alzheimer disease. Thiols are distributed unevenly in cells. Cell surface thiols have been found to serve as part of the structures of proteins related to transporters, receptors, and enzymes. Cell surface thiols are particularly sensitive to exogenous oxidants and reductants. Limited agents are available for imaging cell surface thiols. An agent that can map thiols on the cell surface in live cells will be a valuable tool in understanding how cells respond to exogenous oxidative stress stimuli. TOSBA was designed and synthesized as a thiol-specific and cell surface selective fluorogenic agent. TOSBA effectively imaged cell surface thiols and detected their changes in live NCI-H226 cells. Imaging of thiols on the cell surface by TOSBA was confirmed by fluorescence confocal microscope. TOSBA will be a valuable tool for the study of thiols’ function on the cell surface.
