Pyrophosphates are very important biological anions formed by the hydrolysis of adenosine triphosphate (ATP) into adenosine monophosphate (AMP) in cells. In all organisms from bacteria to humans, inorganic pyrophosphate (PPi) is hydrolyzed to phosphate which in turn helps the DNA, RNA and protein synthesis processes. Alendronate, a PPi analog, is currently being used as a drug for osteoporosis, Paget’s disease, and for complications of prostate cancer. In addition, phosphate anions such as PPi and ATP play key roles in energy<br/>transduction and several major metabolic processes such as DNA replication. The PPi concentrations can provide critical information on these processes and therefore, the detection and quantification of PPi can help identify diseases such as chondrocalcinosis or calcium<br/>pyrophosphate dihydrate (CPPD) crystal deposition disease. We recently showed that, N-Alkyl ammonium resorcinarene chloride, a macrocyclic tetra-salt receptor, binds PPi with a remarkably high binding constant (10 6 M -1 , Figure 1). Herein, we study the effect of the counter anion in the binding of pyrophosphate. The binding processes are studied in solution via NMR spectroscopy, Isothermal Titration Calorimetry (ITC) and UV-Vis/Fluorescence spectroscopy. The results and supported through DFT-based computational calculations.
