Polycyclic guanidinium alkaloids (PGAs) are a class of structurally complex marine natural products which exhibit a broad range of biological activities; however, only little information regarding their biological function and targets are known mainly due to the low natural abundance and their challenging synthesis. Developing a modular and efficient synthetic approach towards PGAs is therefore critical to further develop these scaffolds as chemical probes. Herein, we report a novel synthesis of PGA using bicyclic ß-lactams as key intermediates. From these bicyclic ß-lactams, a short sequence of chemical transformations would allow us to have access to PGA scaffolds, such as the batzelladine and monanchocidin families and open the door to understand their biological activity.
