Engineering receptor or antibody has attracted attention due to the importance to detect various biomolecules and its applications like biosensors using colorimetric assay or electronic signaling with nanomaterials. Antibody has been widely utilized to functionalize nanomaterial surface for development of biosensors due to its high selectivity and binding affinity. However, the complexity of structure and thermodynamically unstable structure has been limitation on using antibody to biosensors. Herein, we newly designed cassette-type artificial antibody that can assembled onto graphene surface for the applications into biosensor by combination of supramolecular self-assembly of coiled-coil and scFv (Single-Chain Fragment Variables). The Fc region of conventional antibody is substituted for heterodimeric coiled-coil protein to enhance thermodynamic structural stability and flexibility that increases biosensor sensitivity. Moreover, the coiled-coil protein is fragmented into three parts by cutting one coiled-coil monomer for the conjugation of pyrene molecule that can interact with graphene conserving the binding ability to target biomolecules. The high thermodynamic stability of self-assembly structure composed of three fragments was measured by both experimental and computational analysis. Our research can suggest novel antibody engineering technique that can be utilized in nanoprobes that can capture specific target biomolecules as well as provide insight into coiled-coil protein structure engineering research field.
