Chlorine is a widely available threat agent that was repeatedly used in alleged attacks in armed conflict globally. A range of chlorinated biomolecules has been suggested as biomarkers of chlorine exposure but no established analytical method is available for unambiguous verification of exposure. Recently, chlorohydrins of phospholipids, were identified as potential biomarkers for chlorine exposure in bronchoalveolar lavage fluid (BALF) of chlorine gas exposed mice.  The markers are chlorination products of unsaturated pulmonary surfactant phospholipids (e.g. phosphatidylglycerols and phosphatidylcholines). In this study, we describe the chemical derivatization of these biomarkers, prior to nano-LC-MS/MS analysis, in order to improve their analytical detection limits. By the improved analytical method, a significant increase of the biomarkers in bronkoalveolar lavage fluid and blood serum samples from exposed animals. In contrast to earlier results on BALF samples, the serum samples of control animals contained a low level of the biomarkers. This result indicate an alternative source of the biomarker in serum samples.<br/>
