The appropriate response to a stimulus within a cell is the result of a coordinated series of changes mediated by cell signaling pathways. Occasionally, members of these pathways are mutated, leading to an inappropriate response to the stimulus. In some cases, this leads to cancer, while in other cases it can cause a developmental syndrome called Noonan Syndrome. My research focuses on explaining why mutations in Ras are found in 30% of cancers, while changes to a related protein, RIT1, may cause Noonan Syndrome. I will focus on comparison of activation and deactivation mechanisms of Ras and RIT1, determining localization and binding interface, and propose reasons that some mutations cause cancer while others lead to Noonan Syndrome.
