Engineered colloidal nanoparticles (NPs) show huge promise as diagnostic, drug delivery, imaging and therapeutic agents. As more engineered NPs become part of our society, it becomes the responsibility of scientists to assess their impacts on living systems. Currently, the experimental focus has been on particle design, since it is well known that the biological interactions of a nanoparticle are a function of its physicochemical properties, such as size, shape, and surface chemistry, but not much focus on the biological system or microenvironments the NPs interacted with. Previous literature have shown proofs that understanding physical interactions of NPs with the microenvironments of various tissues may be better predictors of fate and effects, and our lab have done some related experiments, proving that the NPs indeed interact with the extracellular matrix (ECM) and interference the cell behavior. We hypothesize that NPs in the ECM that surrounds cells alter local concentration gradients of some molecules and thereby influence cell behavior nanoparticles – without the need for the NPs to enter cells. In our experiment, we exame the cell-ECM-NP nexus for the cases where three different types of Au NPs, including poly(sodium-4-styrenesulfonate) (PSS, negatively charged), poly(diallyldimethylammonium chloride (PDADMAC, positively charged) and polyethylene glycol(PEG, neutral) wrapped, are chosen to interact with Monocytic Cell Line THP-1 cells in gradients of Monocyte Chemoattractant Protein-1 (MCP-1) in the media, which has been known to have a clear chemotaxis effect on THP-1 cells. Cell movements are monitored via bright field optical microscopy in chemotaxis µ-Slide. The characterizations of cell migration are analyzed and compared with a series of control experiments. Preliminary results show that PSS-wrapped Au NPs may have effect on MCP-1 guided THP-1 chemotaxis in 3D µ-Slide environment.
