Protein separation materials that are both selective and effective could have wide applications in fields of bioengineering and pharmaceutical industry. In this work, we present a scalable methodology to prepare Hydrophilic cellulose nanofibrous membranes with controlled fiber sizes and functionalized with accessible pyridine rings as chelating group by clean, simple and effective photoinitiation grafting of 4-vinylpyridiene (VP) to cellulose NF surface to form coordinative complexes with metal ions. The chemical structure of grafted cellulose to VP was confirmed by FTIR, contact angle, SEM. Taking advantage of the merits of large specific surface area, highly tortuous open-porous structure, superior chemical stability, and robust mechanical strength, the obtained cellulose-VP-Cu(II) NFM present an excellent integrated lysozyme adsorption performance with a high capacity of 800 mg.g<sup>-1</sup>,  short equilibrium time of 2 h, ease of elution, and good reusability. Smaller fiber size with a higher immobilized metal ion density on membrane surfaces showed greater lysozyme adsorption capacity. The lysozyme adsorption capacity remained consistent during six repeated cycles of adsorption−elution operations with reduction of 5 % of its original adsorption efficiency. Dynamic binding efficiency of 700 mg g-1 can be achieved driven solely by the gravity of protein solution, which matches well with the demands of the high yield and energy conservation in the actual protein purification process. Significantly, the successful synthesis of such intriguing and economic cellulose-VP-Cu(II) NFM from most abundant biopolymer-cellulose-may provide a promising candidate for the next generation of protein adsorbents for rapid, massive, and cost-effective separation and purification of proteins.
