Lipopolysaccharides (LPS) and lipooligosaccharides (LOS) are produced by gram-negative bacteria in its outer membrane. LPS consist of O-antigen, core oligosaccharide and lipid A and LOS is lack of O-antigen. These are important antigenic and immunogenic component and particularly core oligosaccharides are significant. Therefore, these are considered vaccine targets against pathogenic bacteria. In this presentation, we investigated the fragment coupling at the axially oriented 5-position of a 4-O-substituted Kdo acceptor 1with glycosyl donors 2-4and synthesized core trisaccharides of LPS and LOS 5-7[1, 2]. In our strategy, a continuous branched core oligosaccharide 9 that contains a vicinal trans-diol of L-glycero-D-manno-heptose (Hep) and a vicinal cis-diol of 3-deoxy-D-manno-oct-2-ulosonic acid (Kdo) in its structure was first synthesized by using a 3,4-branched Hep donor 8.
