Scientific evidence has shown that western diets have a certain effect on metabolism. Moreover, disorders along with the gut-microbiota-brain axis are closely associated with metabolic diseases. We investigated effect of western diets on metabolic status and neuropeptides of hippocampus in wistar rats under high-fat and high-protein (chicken, pork, casein) diet mode. Proinflammatory factors, neuropeptides and metabolites in hippocampus tissue were detected, and gut microbiota was analyzed by 16S rRNA gene sequencing. The results indicated that communications between gut and brain were modulated in HFHD, by altering structural and diversity of gut microbiota, upregulating proinflammatory factors including IL-6, IL-1β and TNF-α, regulating GABA receptors and BDNF levels, as well as metabolites in hippocampus. Compared with controls, choline, creatine, inositol and N-acetylaspartate were lower in HFHD group, which is correlated with brain memory and cognitive ability impaired. Importantly, HFHD also induced the sign of Alzheimer’s disease, which is characterised by the deposit of brain tau protein and β-amyloid peptide (Aβ) and neuronal loss. Chicken protein diet group showed significant differences from pork and casein protein diet groups. Our findings suggest that western diet could induce metabolic disorders and potentially Alzheimer’s disease via the gut-microbiota-brain axis.
